Method description
Samples are extracted by autoclaving in HCl to breakdown the matrix. A buffer and enzyme are added to convert any thiamin phosphate to thiamin. Alkaline potassium ferricyanide is added to the filtrates to oxidize and convert thiamin to thiochrome. The thiochrome is extracted into iso-butyl alcohol and measured fluorometrically. Blanks are determined by adding NaOH instead of ferricyanide. The thiamin level is determined by comparing the net fluorescence to a standard curve.
