Method description

Samples are extracted by autoclaving in HCl to breakdown the matrix. A buffer and enzyme are added to convert any thiamin phosphate to thiamin. Alkaline potassium ferricyanide is added to the filtrates to oxidize and convert thiamin to thiochrome. The thiochrome is extracted into iso-butyl alcohol and measured fluorometrically. Blanks are determined by adding NaOH instead of ferricyanide. The thiamin level is determined by comparing the net fluorescence to a standard curve.

Information required by submitter

Please supply an estimate of expected thiamine content and indicate the presence of high levels of iron. Samples of vitamin Concentrates should be clearly labeled.

This test is ISO/IEC 17025 accredited through A2LA, Certificate # 2769.01.

High iron content may interfere with the reaction chemistry. Vitamin concentrate results typically have higher-than-normal method variability.
