Method Description:
Samples are incubated with pancreatic a-amylase and amyloglucosidase (AMG) and further treated to remove the nonresistant starch. The resistant starch is then dissolved in KOH, neutralized with acetate buffer, and is quantitatively hydrolyzed to glucose with AMG. Glucose is measured with glucose oxidase/peroxidase reagent (GOP) on a spectrophotometer. The absorbance of this solution at 510 nm is proportional to the resistant starch content of the sample.
Information Required by Submitter:
Please provide estimates for samples containing high (> 10%) resistant starch. The method must be adjusted slightly for samples with high resistant starch.
Additional Information:
This method measures RS2 (granular) and RS3 (retrograded) starches. RS1 starches are not detected by this method. Applicability of this method to RS4 (chemically-cross linked) type starches must be determined on a case-by-case basis.
