Method description

A homogenized sample is weighed and internal standard and phosphate buffer are added. Amylase and protease enzymes are added to further break down the matrix for extraction of the folates. Conjugase is used to de-conjugate the polyglutamate forms of folates to mono-glutamates. The sample is centrifuged, filtered and cleaned using SPE. The folates are analyzed by UPLC-MS/MS. Folic Acid is quantified. Isotopically labeled internal standards are used to control matrix to matrix variation.

Acceptable matrices

All cereal products, flours, milk and soy-based products, infant formulas and baby food composite, vegetables, fruits, protein powder and shake, vitamin concentrates (up to 2 % folates), peanuts, peanut butter and other food matrices.

Unacceptable matrices

Gum, encapsulated vitamins, premixes and supplements above 2%

This test is ISO/IEC 17025 accredited through A2LA, Certificate # 2769.01. This method is not designed for use on vitamin concentrates, and the vitamer 5-methyl tetrahydrofolate will not be quantified.
