Method description

A homogenous sample is dispersed in a phosphate buffer solution at pH 7. Total folates are extracted from the sample using a triple enzyme system. A protease and an amylase are used to digest the food matrix and aid in the release of folates. Rat plasma conjugase is used to hydrolyze the polyglutamate side chain. The assay microorganism, Lactobacillus casei, subsp. rhamnosus (ATCC no. 7469), utilizes total folate derivatives for growth. The sample extract is diluted with an assay medium containing all required growth nutrients for the microorganism except for folates, inoculated with the microorganism, and incubated for 24 hours at 37°C. The growth response of the microorganism (measured as turbidity at 610 nm) in the sample is compared quantitatively to that of known standard solutions of folic acid.

Acceptable matrices

Cereals, fruits, vegetables, flours, grains, bread, pasta, bars, vitamin drink supplements and concentrates that contain added or naturally occurring folates.

Unacceptable matrices

Dairy, meats, eggs, fats and oils, plant based protein, soup, seafood. Premixes/concentrates/supplements above 2%.
