Method description
The sample is completely solubilized by DMSO and fats removed by an alcohol precipitation of the starch. Upon further solubilization and dilution of the sample with acetate buffer, the amylopectin is removed by precipitation with a Concanavilin A. The amount of amylose in the sample is quantified by the conversion of glucose with specific enzymes and measured using a GOPOD reaction. A portion of the sample, prior to the addition of Concanavilin A, is quantified in the same manner yielding the amount of total starch.
Amylose is determined by a ratio of amylose to total starch. Amylopectin is determined by difference, where together they equal 100%.
